After receiving their genre, all six filmmaking teams had one weekend (March 17-19, 2006) to create a short film. All work on the film had to be done during the competition weekend. Each film had to incorporate the character C. Longfellow, an artificial plant as a prop and the line of dialogue, "I know it's around here somewhere." Filmmakers were challenged to create a film approximately 125 seconds long.

Members of the filmmaking teams screened their films at the American Red Cross National Convention in Washington, D.C., after the winners were announced. Team leaders of the winning films received a private mentoring meeting with top Hollywood producers. In addition, the first place team received the first-ever The American Red Cross/48 Hour Film Project Invitational award trophy while the second place (or first runner up film) team took home some cool American Red Cross wear and gear.
Ten members of the American Red Cross National Celebrity Cabinet — Zach Braff, Jamie Lee Curtis, Marcia Gay Harden, Angie Harmon, Marlee Matlin, Julianne Moore, Nancy O’Dell, Elisabeth Rohm, Jane Seymour and Jason Sehorn – graciously agreed to act as judges for the invitational. Each watched and ranked the films, choosing a first place winner.
